肺癌组织中ET-1阳性表达与血管生成关系的研究

摘要 目的:研究内皮素-1(ET-1)在肺癌组织标本中的表达及与肺癌分型、分级的关系,探讨ET-1的表达与微血管密度(MVD)的关系。方法:用免疫组织化学SABC染色方法对不同肿瘤组织进行ET-1表达的测定并行MVD计数,与良性病变组及正常肺组织对照。结果:非小细胞肺癌组织中MVD显著高表达,且与肺癌原发肿瘤大小、淋巴结转移状态和细胞分化程度有密切关系。ET-1在非小细胞肺癌中阳性表达,小细胞肺癌则为阴性表达,两者之间差异具有统计学意义(P=0.008)。非小细胞肺癌ET-1含量与肺癌原发肿瘤大小、淋巴结转移状态和细胞分化程度也有密切关系。ET-1表达阳性组中其MVD显著增高,而ET-1表达阴性的肿瘤组织中MVD仅有微弱表达,两者之间差异有统计学意义(P<0.05)。结论:ET-1在非小细胞肺癌中高表达,且与分化程度和转移状态密切相关;ET-1可能通过肺癌血管生成参与非小细胞肺癌血管发生、发展;ET-1和MVD含量可作为判断非小细胞肺癌分化程度、治疗效果、预后的有效指标。 Objective: To study the relationship between endothehn-1 (ET-1) positive expression and the types of classification of non-small cell lung cancer, observe the connection between ET-1 and microvessel density (MVD) expression and discuss the role of ET-1 in neovascularization of lung cancer. Methods: The expressions of ET-1 and MVD were detected by immuno-histoehemistry. Results: The expression of MVD was notably higher in non-small cell lung cancer. The MVD was closely related to the size of primary cancer, lymph node status and grade of cell differentiation of non small cell lung cancer. The expression of ET-1 was positive in non small cell lung cancer but negative in small cell lung cancer. The positive rate of ET-1 in non small cell lung cancer was significantly higher than that in small-cell lung cancer (P = 0.008). The expression of ET-1 was closely related to the size of primary cancer, lymph node status and grade of cell differentiation of non small cell lung cancer. The expression of MVD was notably higher in ET-1 positive expression tissues. The expression of MVD was weak in tissue with negative expression of ET-1 (P 〈 0.05). Conclusion: There was ET-1 positive expression in non-small cell lung cancer. ET-1 may contribute to neovascularization of non-small cell lung cancer. The expression levels of ET-1 and MVD can be used to detect the grade of cell differentiation, the treatment efficaey and prognosis of non small cell lung cancer.
