白念球菌DNA探针的制备

Preparation of DNA Probe for Candida albicans

应用聚合酶链反应扩增白念珠菌标准株的EO3基因片段，扩增产物125bpDNA用半抗原地高辛标记，制备成探针，该探针显色敏感度达0．01pg，与热带念珠菌、近平滑念珠菌、临床分离的白念珠菌及大肠杆菌、痢疾杆菌、肠球菌等细菌进行斑点杂交试验，结果表明EO3125bpDNA探针仅与白念珠菌杂交，具有高度特异性。

In this study, the EO3 gene of C. albicans was amplified by polymerase chain reaction(PCR). The products,125bp DNA probe, was labeled by hapten digoxigenin, which sensitivity was 0.01pg. The dot-blotting hybridization assay showed that EO3 gene probe only hybridized with C. albicans, but didn't with C. tropicalis, C.paropsilosis and bacteria(E. coli, Shigella, et al). The results showed that the probe was highly specific.
