摘要
目的:研究共轭亚油酸(CLA)在抑制结肠肿瘤发病率,诱导肿瘤细胞死亡,廿烷类、1,2—二酰甘油(DAG)的形成,及对注射1,2一二甲基肼的S-D大鼠的结肠粘膜上的脂肪酸含量中的影响。方法:将120只6周龄的S-D大鼠分为四个喂养组:①藏牦牛脂(TYT);②高原鼢鼠脂(FOPZ);③藏牦牛脂加共轭亚油酸;④高原鼢鼠脂加共轭亚油酸。共轭亚油酸来自青海裸鲤油。四组喂养20周的实验食物,并注射1,2-二甲基肼(20mg/kg),到20周龄时达到180mg/kg。结果:藏牦牛脂+共轭亚油酸喂养组,高原鼢鼠脂+共轭亚油酸喂养组,以及高原鼢鼠脂喂养组的结肠肿瘤发病率低,肿瘤细胞死亡率上升(P<0.05),少量的廿烷类(前列腺素E2和血栓素B2)和结肠粘膜少量的1,2.二酰甘油有加速结肠肿瘤细胞死亡的作用。结论:共轭亚油酸可以增加肿瘤细胞的死亡,具有抗肿瘤的功效,其通过调节结肠粘膜细胞信号传导来诱导肿瘤细胞死亡。
Objective: We researched effects of the CLA on clinical carcinoma incidence, apoptosis, eicosanoid formation, 1,2-diacylglycerol(DAG)and fatty acid profiles in colonic mucosa with 1,2-dimethylhydrazine-injected Sprague-Dawley rats. Methods: One hundred twenty 6-wk-old Sprague-Dawley rats were designed four diet groups. (1)Tiber Yak Tallow(TYT);(2)Fat of Plateau Zoko(FOPZ);(3)TYT+ CLA;(4)FOPZ+CLA. All groups were fed for 20 wk on experimental diets and were injected with 1,2-dimethylhydraxine about 20 wk(for a total dose of 180mg/kg). Results: Rats feeding of TYTC, FOPZC or FOPZ showed reduced incidence of colonic carcinoma,increased apoptotic index values(P〈0.05)and lower levels of eicosanoids(prostaglandin E2 and thromboxane B2)and DAG in colonic mueosa (P〈0. 05). They may share common access of action in promoting the apoptotic process of colon carcinogenesis. Conclusions: All results showed that increased apoptosis by CLA feeding; CLA should have anti-carcinogenic effects that inducing apoptosis through modification of signal transduction in colonic mucosal cells.
