参附注射液保护阿霉素心肌毒性机理研究 被引量:5

摘要 目的:观察参附注射液保护阿霉素心脏毒性的作用机理。方法:急性分离单个心肌细胞,运用TILLVISION钙离子荧光成像及分析系统,观察不同组间单个心肌细胞内游离钙离子的浓度变化。结果:阿霉素可引起心肌细胞内钙离子水平显著升高(P<0.01),参附注射液干预可显著降低异常心肌细胞内游离钙离子浓度;同时,参附注射液还可抑制咖啡因诱导的内钙释放。结论:参附注射液对阿霉素心脏毒性的保护作用可能是通过抑制肌浆网的内钙释放,避免心肌细胞内钙超载,从而维持细胞内钙离子水平,达到保护心肌的作用。 Objective: To observe the mechanism of Shenfu (SF)injection protect myocardial lesion caused by adriamycin. Methods : The single myocardial cells were isolated acutely, free [Ca^2+]i of single myocardial cells were measured by the Tillvision system, to observe the changes of [Ca^2+]i at the different groups. Results: [Ca^2+]i of adriamycin myocardial lesions were increased significantly (P〈0.01) .After using SF, free [Ca^2+]i of myocardial cell caused by adriamycin were decreased significantly, furthermore, the endo-myocardial cell calcium releasing of caffeine induced was inhabited by SF injection, Conclusions : The protective effects of SF injection might inhibit endo-myocardial cell calcium re, leasing of sarcoplasmic reticuium, to advoid calcium overload induced by adriamycin, to achive the effect of protection myocard.
