摘要 目的:优化并确立一套适合视网膜色素上皮细胞二维电泳的实验方法。方法:提取视网膜色素上皮细胞可溶性蛋白,改进条件以确立对视网膜色素上皮细胞蛋白进行二维电泳的实验流程。结果:该实验条件对视网膜色素上皮细胞蛋白达到了良好的分离效果,并能通过分析找出正常与病变细胞蛋白表达差异。结论:该优化的实验流程能有效分离视网膜色素上皮细胞蛋白质,并为进一步的差异蛋白质组研究奠定基础。 Objective: To optimize and establish two-dimensional electrophoresis(2-DE) steps for proteome analysis of retinal pigment epithelium(RPE) cells. Methods: Total soluble proteins of ARPE-19 cells were extracted, then experimental conditions were changed to find out the best one for RPE. Results: There were 300± 10 spots apparently displayed, and differential expressing proteins can be found out after image analyzing of 2-DE gels between the experiment group and the normal control group. Conclusion: These 2-DE steps meet the qualifications for research in RPE cells proteome and are useful to further study on differential proteome profiles.
