摘要
目的:观察安宫清开灵注射液对胶原酶诱导的自发性高血压大鼠(SHR)脑出血模型脑组织中前炎症细胞因子TNF-α、IL-1β以及血清中黏附分子sICAM-1含量的影响,探讨安宫清开灵注射液阻抑脑出血后炎症反应的作用环节以及作用效果。方法:基底核注射胶原酶法复制大鼠脑出血模型;应用放射免疫法检测脑组织匀浆中TNF-α和IL-1β含量的变化,酶联免疫吸附方法检测血清中sICAM-1含量。结果:脑出血48h和7d,大鼠脑组织匀浆中TNF-α和IL-1β的含量以及血清中sI-CAM-1含量均显著升高;安宫清开灵注射液各剂量组脑内TNF-α和IL-1β的含量以及血清中sI-CAM-1含量显著低于模型组(P<0.01或P<0.05)。结论:安宫清开灵注射液可以显著抑制前炎症细胞因子TNF-α和IL-1β的生成,以脑出血急性期(出血后24h)的抑制作用较突出,也可以阻抑黏附分子sICAM-1的合成与分泌,并随着用药剂量增加而效应增强的剂量依赖关系。可见安宫清开灵注射液对炎症反应的启动具有显著的阻抑作用。
This paper studies the effects of An Gong Qing Kai Ling Injection on the level of TNF - α, IL - 1 β in the brain tissue, and sICAM - 1 in the serum of SHRs having an intracerebral hemorrhage, in an attempt to understand the links and effects of An Gong Qing Kai Ling Injection in repressing inflammatory reactions. In the study, SHRs were randomly divided into six groups: normal, model, low-dose treatment, middle -dose, high -dose, and positive control. Intracerebral hemorrhage was induced in SHRs using collagenase VII. RIA (radioimmunoassay) was employed to measure the concentration of TNF - α and IL - 1 β in SHRs' brain tissues, following the attack of cerebral hemorrhage, sICAM - 1 determination was made using an ELISA (enzyme - linked immunosorbent assay) , in line with the manufacturer' s instruction. Results showed that after 48h and 7 days of intracerebral hemorrhage induction, the median TNF -α, IL- 1β and sICAM - 1 levels in the model was higher than that of control (P 〈0.01 ). The median TNF - α, IL - 1β and sICAM - 1 levels in the treated groups was lower than that of model (P 〈 0.01 or P 〈 0.05 ). The median TNF - α, IL - 1β and sICAM - 1 levels in the high - dose group was lower than that of the positive group (P 〈 0.01 ). It is found that An Gong Qing Kai Ling Injection is of a noticeable effect on reducing the level of inflammatory factors, especially in the 48 hours after the onset. In addition, it cuts down the level of sICAM - 1 in a dose - dependent manner. It is concluded that An Gong Qing Kai Ling Injection has a noticeable effect in preventing the onset of inflammation, allowing efficient regulation of TNF - α without eliminating the constitutive levels that are required for the cell survival.
