摘要
本文报道以本室自制的梳状硝酸纤维素滤膜为载体,采用问接Dot—ElISA法检测患者血清抗HBc IgM,IgG,IgA。研究表明,本法除可在2h内快速获得结果外,尚有特异性强,敏感性高(检测抗HBc IgG可达0.4ng/ml),操作简便,结果易于观察等优点。与板上ELISA法进行比较检测,符合率为94%。在检测抗HBc IgM时,不受RF因子的影响。将本法所用材料组装成新型试剂盒后,在4℃下可保存半年以上,便于基层单位应用。
This paper described an indirect method of Dot-ELISA for the detection of Anti-HBc specific IgG, lgM and lgA. The eomb-like nitrocellulese filter was used as solid carrier. Through blocking, neutralizing, cross-reacting and 2-ME destroying experiment, we demonstrated that the serum antibodies detected by this method were the anti-HBc specific immunoglobulins. We compared the method with plastic plate ELISA, the coincidence rates were 94% for Anti-HBc IGG, 97% for Anti-HBc IgM and 95% for Anti-HBc lgA, respectively. The sensitivity of this methed for detecting HBc specifie IgG was 0.4ng/ml. It was shown that this method was specific, sensitive and rapid. The whole experiment could be completed within 2 hours. The result was very easy to read, and we didn't find any interferrenee from RF factor, when detecting Anti-HBc IgM. The kits developed in our laboratories could be stored at 4℃ for more than half a year, so it should be widely used in ordinary laboratories.
