Determination of total polyphenol in barley seed extracts by Folin-Ciocalteu colorimetry

摘要 采用Folin-Ciocalteu(FC)比色法测定大麦提取液中总多酚的含量,确定以没食子酸为标准的最佳反应条件。结果表明,FC试剂浓度为0.2mol/L,Na2CO3溶液浓度为15%,在室温下反应60min,浓度在1~9μg/mL时与其吸光值有良好线性关系。方法操作简单、结果重现性好,可用来测定大麦提取物中总多酚的含量。 The total polyphenol(TP) content of barley extracts was analyzed by Folin-Ciocalteu colorimetry. The optimum conditions were investigated using gallic acid as standard. The results showed that when the concentration of Folin-Ciocalteu reagent and Na2CO3 were 0.2mol/L and 15%, reaction 60min in room temperature, there was a good linearity between the absorbance and gallic acid concentration from 1-9μg/mL in 760nm. The method was simple, accurate and suggested as an appropriate method to measure the TP content in barley extracts.
