血管紧张素Ⅱ1型受体A1166C多态性与唐山地区原发性高血压 被引量:2

摘要 目的:探讨血管紧张素Ⅱ1型受体(AT1R)A1166C多态性与唐山地区原发性高血压(EH)的关系。方法:入选研究对象581例,其中正常对照人群291例,EH患者290例。所有研究对象用常规方法提取白细胞DNA,采用多聚酶链反应结合限制性内切酶(DdeI)方法检测AT1R基因A1166C多态性。结果:对照组和EH组CC、AC和AA基因型频率为0.4%,18.2%,81.4%;0%,12.1%,87.9%。对照组和EH组A等位基因频率分别90.5%和94.0%。EH组AT1R基因A1166C多态性AA基因型和A等位基因频率均较对照组增多,差异有统计学意义(P均=0.04)。结论:AT1R基因A1166C多态性与唐山地区EH有关,可能是EH的一个遗传标志。 Objective: Renin-angiotensin-aldosterone system (RAAS) is important in the development of essential hypertension. To investigate the relationship between angiotensin Ⅱ type Ⅰ receptor(AT1R) gene A1166C polymorphism and essential hypertension in Tangshan district. Method:291 healthy people were enrolled into this study as control group.290 patients with essential hypertension were enrolled into this study. DNA was extracted from white blood cell. Segments of AT1R were amplified from DNA by polymerase chain reaction(PCR). PCR products were restricted with 5 U of DdeI for 3 hours at 37℃. The digested products were subjected to electrophoresis in 1.5% agarose gels, and stained with ethidium bromide. Result:AT1R gene Al166C polymorphism genotype in the cases and controls were AA,AC and CC 87.9% ,12.1% ,0% ; 81.4% ,18.2% ,0.4% respectively. A allele frequencies were 94.0% and 90.5% respectively. The frequency of AA genotype and A allele was significantly higher than that in the control(P = 0.04,0.04). Conclusion: the Al166C polymorphism in the AT1R gene is associated with essential hypertension in Tangshan district, A allele maybe a genetic marker in essential hypertension.
