三种中药复方对脑缺血再灌注大鼠脑组织ICAM-1和NF-κB表达的动态影响 被引量:4

摘要 目的比较三种中药复方(益气活血汤、镇肝息风汤、星蒌承气汤)对脑缺血再灌注大鼠脑组织I-CAM-1和NF-κB蛋白表达的动态影响,探讨其抗脑缺血再灌注炎性损伤的机制。方法采用线栓法大脑中动脉制作大鼠局灶性脑缺血再灌注模型,脑缺血2h,分别再灌注24、48和72h。大鼠随机分为假手术组、模型组、益气活血汤组、镇肝息风汤组和星蒌承气汤组。采用免疫组化SABC染色法分别检测缺血半暗带ICAM-1和NF-κB蛋白表达。结果脑缺血2h再灌注各时间点与假手术组比较,ICAM-1和NF-κB蛋白表达阳性细胞数显著增加;再灌注24h各治疗组与模型组比较,ICAM-1蛋白表达阳性细胞数显著减少;再灌注48h各治疗组NF-κB蛋白表达阳性细胞数显著降低,ICAM-1仅星蒌承气汤组显著降低;再灌注72hICAM-1和NF-κB蛋白表达阳性细胞数均显著减少;组间比较24h镇肝息风汤组优于其他组,48h星蒌承气汤组优于其他组,48h益气活血汤组优于其他组。结论上述三种中药复方可以通过降低ICAM-1和NF-κB蛋白表达抗脑缺血再灌注炎性损伤。 Objective: Comparing the influence of 3 Chinese herbal compound formula on the expression of ICAM - 1 and NF - κB protein in cerebral tissue after focal cerebral ischemia - reperfusion in rats, to investigate the mechanism of those in resisting on localcerebral ischemia and reperfusion inflammatory injury. Methods: The local cerebral ischemia-reperfusion models were established by intraluminal thread occlusion of the middle cerebral arteries (MCAO) . The middle cerebral arteries of rats were occluded for 2 hours and reperfused for 24, 48 and 72 hours individely. The test animals were divided into the Sham group, the model group, Yiqihuoxue group, Zhengan xifeng group and Xinglouchengqi group. ICAM - 1 and NF - κB protein expression were detected by immunohistochemistry method in ischemic penumbra(IP) . Results: The positive cell number of ICAM - 1 and NF - κB protein expression were higher than those in the Sham group in MCAO rats. The positive cell number of ICAM - 1 protein expression in all treatment groups for 24h and 72h were higher than those in the model group, only Xinglouchengqi group for 48h. The positive cell number of NF - κB protein expression in all treatment groups for 48h and 72h were higher than those in the model group. Therapeutic effect for 24h in Zhengganxifeng group was superior to that in other groups (NF - κB), Xinglouchengqi group, for 48h was superior to that in other groups(ICAM - 1 )and Yiqihuoxue group for 48h was superior to that in other groups. Conclusion: The former, 3 Chinese herbal compound formula could resist pathologic inflammatory injury after focal cerebral ischemia and reperfusion in MCAO rats by reduced expression of ICAM - 1 and NF - κB protein.
