摘要
目的测定不同种类、不同产地的细辛根茎、叶柄、叶中马兜铃酸及挥发油含量,指导临床用药。方法采用高效液相色谱法(HPLC法)测定马兜铃酸含量,色谱柱为phenomenex Luna C18柱(250mm×4.60mm,5μm),流动相为乙腈-磷酸二氢钠(39∶61)(0.05mol/L),检测波长为387nm,流速为1.00mL/min。挥发油按2000年版《中国药典(一部)》中附录ⅩD甲法测定。结果马兜铃酸进样量在5.628~37.52ng范围内与峰面积线性关系良好,r=0.9998,回归方程为Y=1305248.34X-264.16,平均加样回收率为97.39%,重现性试验的RSD为1.28%。结论按细辛部位,挥发油含量根茎>叶>叶柄,马兜铃酸含量根茎<叶柄<叶。
Objective To develop a HPLC method for the determination of aristcloch acid and naphtha in asarum. Methods The analysis was carried out using Phenomenex luna C18 (250 mm×4.6 mm, 5 μm).The mobile phase consisted of acetonitrile-0.05 mol/L NaH2PO4 (39:61). The wavelength of detection was at 387 nm. The naphtha analysis was carried out by the method in Chinese Pharmacopoeia (2000 edition). Results Aristolochic acid showed a good linear relationship at the rang of 5. 628-37.52 ng. The regression equation was Y= 1 305 248. 34X-264.16( r=0. 999 8), and RSD was 1.28%. The average recovery rate of aristolochic acid was 97.39%. Conclusion The naphtha content of asarum is radix 〉 leaf 〉 leafstalk. The aristolochic acid content is radix 〈 leafstalk 〈 leaf.
