摘要
[目的]为了解2006年引起一起风疹暴发疫情的风疹病毒的分子生物学特征,确定其基因型别。[方法]用Vero-Slam细胞从风疹暴发患者的标本中分离风疹病毒,通过逆转录-聚合酶链反应(RT-PCR)和序列测定与分析鉴定其基因型。[结果]四川省首次分离出的2株风疹野病毒全部鉴定为2B基因型。与3株WHO风疹病毒2B基因型参考株(I11-IS-68、TS34-CH-00和TAN-IND-00)核苷酸同源性分别为93.6%、95.6%和97.1%;氨基酸同源性分别为99.5%,99.5%和100%。[结论]此次疫情是由2B基因型风疹病毒引起的暴发疫情,此项研究为将来四川省风疹病毒的分子流行病学研究打下了基础。
[Objective] To understand genetic characterization and genotypes of wild-type rubella viruses in an rubella outbreak in 2006. [ Methods] Rubella viruese were isolated from specimens on Vero-Slam cells, reverse-transcription polymerase chain reaction (RT-PCR) and sequencing were followed to identify the genotypes. [ Results] 2 strains of wild-type rubelld virus isolated in Sichuan were genotype 2B. By comparison with 3 other genotype 2B reference isolates (III-IS-68. TS34-CH-00 and TAN-IND-00), the identities of nucleotides were 93.6%, 95.6% and 97.1% respectively, and the identities of anino acids were 99.5%, 99.5%, and 100% respectively. [Conclusion] This outhreak of rubella is caused by genotype 2B rubella virus. The research will lay a foundation for rubella virus molecular epidemiology in Sichuan.
