摘要
目的观察MMC处理的外周血单核细胞(PBMCs)对大鼠同种异体心脏移植的影响。方法BN大鼠作为供体,Lewis大鼠作为受体,体重在200~300g。PBMCs于含有MMC的细胞培养液中培养30min后于移植前1周静脉注射入受体,然后接受心脏移植,观察移植心脏的存活情况;并利用FACS检测MMC对PBMCs存活率的影响。结果MMC处理过的PBMCs诱导了细胞的凋亡;接受MMC处理过的PBMC的大鼠,移植心脏的存活时间明显得以延长。结论MMC处理的PBMCs可以延长大鼠移植心脏的存活时间,对移植心脏的延长作用可能是由于MMC诱导了细胞的凋亡引起的。
Objective To study the effect of MMC-treated PBMCs on the survival of heart transplantation in rats. Methods BN and Lewis rats, weighing from 200-300 g, served as donors and recipients respectively. PBMCs were cultured in MMC containing medium for 30 rain, and naive or MMC-treated PBMCs were used to treat the recipient i. v one week before heart transplantation. The function of the graft heart was monitored daily by direct palpation through abdominal wall. And the dead rate of MMC-treated PBMCs was measured by FACS in order to know the effect of MMC on the survival of naive PBMCs. Resuits The apoptosis was induced in the MMC-treated PBMCs. The survival of the graft heart was significantly prolonged with the use of MMC treated PBMCs as compared with that of untreated PBMCs treated recipient rats (8.6 vs 18.2 days). Conclusion MMC-treated PBMCs can prolong the survival of graft heart in rats, which might be contributed to the apoptosis of PBMCs induced by MMC treatment.
