摘要
目的:观察不同方式损伤兔坐骨神经电后受损区域神经许旺细胞Kv1.5钾通道基因的表达水平。方法:随机将实验动物分为3组:50V电损伤组、100V电损伤组、神经切割伤组,双酶消化法分离受损区域坐骨神经许旺细胞并体外培养,应用逆转录-聚合酶链反应(RT-PCR)技术比较各组许旺细胞Kv1.5 mRNA的表达水平。结果:3组动物损伤区域神经许旺细胞均有Kv1.5基因的表达,其中损伤后3~7dKv1.5基因的表达最为显著;50V组及神经切割伤组两组之间比较无显著性差异(P>0.05);而100V组Kv1.5基因的表达水平较低,与其他两组有显著性差异(P<0.05)。结论:坐骨神经损伤后许旺细胞Kv1.5基因的表达与损伤后时间及损伤电压大小有关,可能与许旺细胞的增殖有密切关系。
Objective: To study the difference of expression of Kv1. 5 mRNA of Schwann cell in rabbit sciatic nerve injured with different methods. Methods: The experimental animals were randomized into 3 groups: 50V electrical injury group, 100V electrical injury group and nerve concis group, Schwann cells were separated from the region injured with trypsin and collagenase and then cultured in vitro. The difference of expression of Kvl. 5 mRNA of Schwann cell in all group.s we,'e compared by reverse transcription polymerase chain reaction. Results : Kv1. 5 mRNA of Schwann cell was expressed in all groups and was more significant from 3 day to 7 day. There was no obvious difference l)etween 50V electrical injury group and nerve concis group(P〉 0. 05). The expression of Kvl. 5 mRNA of Schwann ceil in 100V electrical injury group was low (P〈 0.05). Conclusion, The expression of Kv1. 5 mRNA in sciatic nerve Schwann cells is concerned with the time after injury and injuring electrical voltage, which may have relationship with proliferation of the Schwann ceil after injury.
