摘要
选取鸡源补体C3α链的21个氨基酸残基序列,人工合成模拟C3分子的21肽[741-761],与牛血清白蛋白载体偶联结合后,制成人工合成抗原复合物。将此人工合成抗原加弗氏佐剂制成油乳剂疫苗,免疫注射家兔制备成抗21肽抗体。从抗血清中提纯IgG,用以检测不同动物血清中C3含量。结果表明,人工合成的C3分子的21肽复合抗原具有很强的免疫原性,制成的抗血清的IgG效价达29,且能与不同动物血液的C3分子发生特异性结合;用该IgG对鸡、猪、山羊、牛和豚鼠体内的C3进行了检测,其含量分别为(0.34±0.005)、(1.47±0.008)、(1.70±0.005)、(1.78±0.007)、(4.18±0.008)g/L,发现鸡体内的C3含量明显低于其他几种动物,差异极显著(P<0.01),这可能是鸡体对细菌性疫苗免疫效果不佳的主要原因。
The 21 amino acid of chicken complement C3^741-761 was selected out of its a chain to synthesize the 21 peptide. The 21 peptide was coupled with the cattle serum albumin to fabricate the antigen compound. Which mixed with the Frund adjuvant and emulsificated to immunise the rabbit to get the anti-21 peptide antibody. The IgG was purified to assay the C3 content in the serum of different animals. It showed that the antigenicity of the synthesized antigen is very strong, The efficacy of purified IgG from the anti-serum is 2^9. The antibody can bind tothe complement C3 of different animal. The C3 content of different animal serum assayed by the antibody are : chicken (0.34±0.005)g/L, pig(1.47±0.008)g/L, goat(1.70±0.005) g/L, cattle(1.78±0.007) g/L, guineapig(4.18±0.008)g/L. It showed that the C3 content of the chicken is extremely lower than the other animal (P〈 0.01). This may account for the less effects of the bacterial vaccines in chicken.
