摘要
目的研究VEGF和COX-2在喉鳞状细胞癌中的表达,探讨它们与喉癌的生长、浸润及转移的关系。方法应用RI-PCR方法检测62例喉鳞状细胞癌、54例癌旁组织和9例正常喉黏膜组织中的VEGF mRNA及COX-2mRNA的表达。结果VEGF mRNA在喉鳞癌组织中高表达,在正常喉黏膜中低表达,差异有显著性(P<0.01),并且在喉鳞癌组织Ⅰ期+Ⅱ期与Ⅲ期+Ⅳ期表达之间差异有显著性(t=-6.279,P<0.01)。COX-2mRNA在喉鳞癌组织中高表达,在正常喉黏膜中不表达,差异有显著性(P<0.01),并且在喉鳞癌组织Ⅰ期+Ⅱ期与Ⅲ期+Ⅳ期表达之间差异有显著性(t=-9.559,P<0.01)。在喉鳞癌中VEGF mRNA和COX-2mRNA的表达呈正相关,相关系数r=0.756,P<0.01。结论VEGF和COX-2与喉鳞癌的生长、浸润及转移有密切关系并有协同作用。
Objective To study the expressions of vascular endothelial growth factor (VEGF) and cyclooxygenase--2 (COX-2) in human laryngeal squamous cell carcinoma and its significance. Methods In this study, we investigated the expression of VEGF mRNA and COX-2 mRNA in sixty-two human laryngeal squamous cell carcinoma and fifty-four tissues adjacent to tumor and nine normal human laryngeal mucous membrane using techniques of semi-quantify RT-PCR. Results It was showed that the expression level of VEGF mRNA was significantly increased in human laryngeal squamous cell carcinoma as compared with the normal human laryngeal mucous membrane (P〈0.01), and showed that the expression level of COX-2 mRNA was significantly increased in human laryngeal squamous cell carcinoma as compared with the normal human laryngeal mucous membrane (P〈0.01), and that the expression level of VEGF mRNA and COX-2 mRNA were significantly increased in Ⅱ and Ⅳstage tissues of human laryngeal squamous cell carcinoma as compared with the Ⅰ and Ⅱ stage tissues of human laryngeal squamous cell carcinoma (P〈0.01). There was a high positive correlation between VEGF and COX-2 expression in human laryngeal squamous cell carcinoma (r=0,756, P〈0.01). Conclusions These data raise the possibility that VEGF and COX-2 many play key roles in the growth, invasion and metastasis of human laryngeal squamons cell carcinoma.
