摘要 目的:肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)对体外培养的人牙囊细胞(dental follicle cells,DFC)表达骨保护素(osteoprotegerin,OPG)的影响,探讨TNF-α在牙齿萌出过程中的作用。方法:原代培养人牙囊细胞;取3-6代细胞,培养基中加入TNF-α,在不同浓度和时间点,MTT检测细胞增殖,RT-PCR检测OPG mRNA的表达。结果:TNF-α在10-50 ng/mL促进人牙囊细胞的增殖,TNF-α与牙囊细胞共孵育,轻度降低OPG的mRNA的表达。结论:TNF-α在体外培养的牙囊细胞有降低OPG表达的作用。 AIM: To study the effects of tumor necrosis factor - 1 α (TNF - α) on the expression of osteoprotegerin (OPG) in human dental follicle cells. METHODS: The culture of human dental follicle cells was established , and the third to sixth passage cells were incubated with TNF - α for both time and dose dependent studies. MTT assays were used to evaluate the cells proliferation. Reverse transcriptase- polymerase chain reaction (RT- PCR ) were used to measure the amount of OPG mRNA in the conditioned mediums. RESULTS: This study showed that the proliferation activity increased significantly by dental follicle cells after incubation with 10 - 50 ng/mL TNF - α at third day. The expression of OPG slightly reduced after incubation of the human dental follicle cells with 10 ng/mL TNF - α at 1 h. CONCLUSIONS : TNF - α can slightly reduced the expression of OPG in human dental follicle cells.
