IL-1β mRNA和TNFα mRNA在慢性根尖周病损组织中的表达

Expression of IL-1β mRNA and TNFα mRNA in apical periodontitis

目的:检测白细胞介素1β(interleukin-1 beta,IL-1β)mRNA和肿瘤坏死因子α(tumornecrosis factor alpha,TNFα)mRNA在慢性根尖周病损中的表达以及它们与病理学表现之间的关系,探讨IL-1β和TNFα在慢性根尖周炎发病中的作用。方法:采用RT-PCR法,从mRNA水平检测IL-1β及TNFα在10例慢性根尖周肉芽肿及3例根尖周囊肿中的表达,3例因阻生拔除的健康第三磨牙牙周膜组织作为对照,分析IL-1βmRNA和TNFαmRNA的表达与慢性根尖周病损的病理学表现(包括病损的病理分型、炎症细胞浸润程度)之间的关系。结果:IL-1βmRNA在根尖周肉芽肿病损组明显增高,表现为与β-actin水平的比值(1.36±0.12)增高,在根尖周囊肿病损组(0.87±0.30)较对照组(0.64±0.02)有增高的趋势,但统计学分析无显著性差异。IL-1βmRNA在轻、重度炎症细胞浸润组较中度炎症细胞浸润组明显增高(P<0.05)。TNFαmRNA水平在根尖周肉芽肿组和根尖周囊肿组均显著高于对照组(P<0.05),但两组间并无显著性差异。TNFαmRNA水平在轻、中、重度炎症细胞浸润组之间也无显著性差异。IL-1βmRNA和TNFαmRNA水平未发现有明显的相关关系。IL-1βmRNA和TNFαmRNA水平与病损大小无关。结论:IL-1β和TNFα在慢性根尖周炎的发病中具有重要的作用,但也存在其他细胞因子的协同作用。

AIM：：To quantify interleukin - 1 beta （IL - 1β） mRNA and tumor necrosis factor alpha （TNFα） mRNA in lesions of chronical apical periodontitis and detect the relation to the histology of the involved tissue. METHODS：A total of 13 periapical lesions including 10 periapical granulomas and 3 periapical cysts were selected. Normal periodontal ligament tissue from extracted third molors were used as control. The expression of IL - 1β and TNFα mRNA were detected by reverse transcription polymerase chain reaction （ RT - PCR ） technique. The relationship between the expression of those cytokins and histological findings （ including the type of histology, the degree of inflammatory infiltrates） was analyzed. RESULTS：A significant higher level of IL- 1β mRNA was found in periapical granulomas （ 1.36± 0. 12） , though the mean relative amount of IL - 1β mRNA in periapical cysts （0.87 ±0.30） was lower than that of periapical granulomas, but higher than that of the normal control （ 0.64 ± 0.02 ）. The relative amount of IL - 1β mRNA in the group with light and heavy degree of inflammatory cells infiltration was higher than that of moderate degree of inflammatory cells infiltration. A significant higher level of TNFα mRNA was found in periapical granulomas and periapical cysts. CONCLUSION：IL- 1β and TNFα might play more important roles in the pathoge nesis and development of periapical lesions.
