白细胞介素6基因型及血清水平与食管癌的相关性研究 被引量:10

Serum level and genotype of interleukin-6 in patients with esophageal cancer

摘要 目的了解食管癌患者白细胞介素6(IL-6)基因启动子-572C/G、-634 C/G 的多态性,探讨 IL-6的基因型及其血清水平与食管癌的关系。方法采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法,检测118例食管癌患者和130例健康对照者 IL-6基因多态性,同时采用酶联免疫吸附试验检测 IL-6的血清水平。结果 IL-6-634C/G 基因型和等位基因频率在食管癌组和对照组比较,差异有统计学意义(均 P<0.05);与对照组相比,G 等位基因携带者患食管癌的相对风险是 C 等位基因的1.759倍(OR=1.759,95%可信区间为1.150~2.691)。食管癌组IL-6水平显著高于对照组(16.9 ng/L±5.3 ng/L vs 4.6 ng/L±2.6 ng/L,P<0.01),携带 G 等位基因的食管癌患者血清 IL-6水平显著高于不携带者(18.8 ng/L±6.1 ng/,L vs 13.2 ng/L±6.0 ng/L,P<0.01)。IL-6基因-572C/G 多态性在食管癌组和对照组间比较差异无统计学意义(P>0.05)。结论 IL-6基因-634C/G 多态性与食管癌之间可能存在相关关系;携带 G 等位基因的个体可能通过促进 IL-6的高度表达进而增加了食管癌的发病风险。 Objective To investigate the polymorphism of the interleukin-6 (IL-6) gene promoter- 572C/G and -634C/G in patients with esophageal cancer, and to study the relation between the serum level and genotype of interleukin-6 and esophageal cancer. Methods Peripheral blood samples were collected from 118 patients with esophageal cancer and 130 healthy persons as controls. The polymorphism of IL-6 was detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. The serum level of IL-6 was determined by enzyme-linked immunosorbent assay ( ELISA ). Results The frequency of the genotype GG of the IL-6 (-634) site in the cancer group was 12.7%, significantly higher that that of the control group (3.8%, P 〈0. 05). The risk of esophageal cancer of the G allele carriers was 1. 759 times that of the C allele carriers ( OR = 1. 759, 95% CI = 1. 150-2. 691 ). The serum level of IL-6 of the esophageal cancer group was ( 16.9 ± 5.3 ) ng/L, significantly higher than that of the control group [ (4.6±2.6) ng/L, P 〈 0.01 ]. The serum level of IL-6 of the esophageal cancer with the G allele carriers was ( 18.8±6.1 ) ng/L, significantly higher than that of the esophageal cancer without the G allele carriers [ (13.2±6.0) ng/L, P 〈 0.013. There was no significant difference in the distribution of the IL-6 gene- 572C/G polymorphism between the 2 groups (P 〉 0.05) . Conclusion IL-6 gene-634C/G polymorphism is associated with the esophageal cancer. The IL-6 allele G carriers may be at increased risk of the esophageal cancer because of the increase of the IL-6 expression.
