The effect of hydroxyethyl starch_(130/0.4) on expressions of GPⅡb/Ⅲa and CD62P of the platelet membrane

摘要 目的观察输注羟乙基淀粉130/0·4(HES130/0·4)对血小板膜糖蛋白(GP)Ⅱb/Ⅲa和CD62P的影响。方法ASAⅠ~Ⅱ级择期手术患者30例,随机分为HES130/0·4(V)组和复方乳酸钠(R)组,每组15例,均在手术前以15ml/kg的速率输入。于输注前(T1)、输注后15min(T2)和60min(T3)采取静脉血,用流式细胞术测定GPⅡb/Ⅲa和CD62P的表达量。结果V组T2时间点的GPⅡb/Ⅲa和CD62P的表达水平较T1有显著性降低(P<0·05);T3时间点的GPⅡb/Ⅲa和CD62P的表达水平与T1比较无显著性差异(P>0·05)。结论HES130/0·4术前行急性高容量血液稀释时,短时间内可抑制对GPⅡb/Ⅲa和CD62P的表达,对凝血功能影响轻微。 Objective To investigate the effects of hydroxyethyl starch130/0.4 ( HES130/0.4 ) on the expressions of platelet glycoprotein Ⅱb/Ⅲa(GPⅡb/Ⅲa) and CD62P. Methods Thirty patients (ASA class Ⅰ or Ⅱ) scheduled for selective surgery were randomly divided into two groups with 15 cases each. Hydroxyethyl starch group(V) and Ringer's lacate solution group(R) were infused HFS130/0.4 and Ringer's lacate solution, respectively, in a dose of 15 ml/kg before operation. Venous blood samples were taken before infusion(T1 ), at 15 min(T2 ) and 60 min(T3 ) after infusion for deterruination of the percentage of GP Ⅱb/Ⅲa and CD62P positive platelet using flow cytometry. Results GPⅡb/Ⅲa and CD62P expressions of group V at T2 were significantly lower than those at T1 (P〈 0. 05). Conclusion Acute hypervolemic hemodilution (AHH) with HES130/0.4 before surgery can inhibit the expression of platelet membrane GP Ⅱb/Ⅲa and CD62P. There is a slight effect of infusion HES130/0.4 on the blood coagulation function.
