中国南方汉族人群不稳定性心绞痛患者基质金属蛋白酶9基因C1562T多态性的特点 被引量:5

摘要 目的:了解基质金属蛋白酶9基因C1562T多态性与中国南方汉族人群不稳定型心绞痛易感性的关系。方法:选择2003-06/2006-06湘南学院附属医院对经冠状动脉造影证实的不稳定型心绞痛患者90例(不稳定型心绞痛组)和同期冠状动脉造影阴性、排除冠心病诊断的患者70例(对照组)。采用酶联免疫吸附试验测定血浆基质金属蛋白酶9水平,采用多聚酶链反应-限制性内切酶片断长度多态性技术分析基质金属蛋白酶9基因中C1562T基因多态性,比较各组的基因型和等位基因频率。结果:所有受试者完成测试并进入结果分析,无脱落者,DNA抽提均成功。①不稳定型心绞痛组血浆基质金属蛋白酶9水平明显高于对照组[(108.65±32.54),(51.12±12.47)μg/L](P<0.01)。②不稳定型心绞痛组基质金属蛋白酶9基因CT基因型频率(28%)以及T等位基因频率(16.1%)均高于对照组(16%,9.3%),差异有显著性(P<0.05)。③C1562T位点CT/TT基因型患者血浆基质金属蛋白酶9水平高于CC基因型患者[(92.12±39.56),(73.46±28.67)μg/L](P<0.05)。结论:基质金属蛋白酶9基因C1562T多态性与中国南方汉族人群不稳定型心绞痛有关,1562T等位基因可能是不稳定型心绞痛遗传易感性的基因标记之一。 AIM: To investigate the association between the matrix metalloproteinase-9 (MMP-9) gene C1562T polymorphism and the susceptibility to unstable angina pectoris (UAP) in Han population of South China. METHODS: Ninety patients with arteriographically documented UAP and 70 cases without coronary heart disease proved by coronary arteriography were selected as angina group and control group, respectively from the affiliated Hospital of Xiangnan College between June 2003 and June 2006. Plasma levels of MMP-9 were measured by enzyme-linked immunosorbent assay (ELISA). Promoter C1562T gene polymorphism of MMP-9 was analyzed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Genotype and allele frequency were compared between the two groups. RESULTS: All subjects completed the examination and entered the result analysis without any loss. DNA extraction was successful. (1)Plasma MMP-9 level of the angina group was significantly higher than that of the control group [(108.65±32.54), (51.12±12.47) μg/L, P 〈 0.01]. (2)The frequency of CT genotype (27.8%) and T allele (16.1%) in the angina group were higher compared with those in the control group (P 〈 0.05). (3)Plasma MMP-9 level of CT/TT genotype patients was higher than that of CC genotype patients [(92.12±39.56), (73.46±28.67)μg/L, P 〈 0.05]. CONCLUSION: The genetic polymorphism in MMP-9 promoter (C1562T) is associated with the unstable angina pectoris in Han population of South China. 1562T allele may be one of the genetic marks of the susceptibility to unstable angina pectods.
