促红细胞生成素对大鼠机体铁状态及腹腔巨噬细胞铁代谢的影响 被引量:4

摘要 网状内皮系统(reticuloendothelial system,RES)在维持机体铁稳态方面起着重要作用.但其调控机制尚不十分清楚.通过皮下注射重组人促红细胞生成素(recombinant human erythropoietin, rHuEPO)引起大鼠红细胞的生成显著增加,改变了血清转铁蛋白饱和度以及机体铁储存状态.利用RT-PCR及Western blot检测发现腹腔巨噬细胞二价金属离子转运蛋白(duodenal divalent metal trans- porter 1,DMT1)的表达显著下降,膜铁转运蛋白1(ferroportin 1,FPN1)的表达显著上升.同时采用^(55)Fe同位素标记示踪法显示腹腔巨噬细胞的铁摄取减少,铁释放量显著增加.研究结果表明在红细胞生成过程中,巨噬细胞摄铁的降低,可能是由于DMT1表达减少所致,巨噬细胞铁释放的增加,可能是由于FPN1表达上升弓J起,肝脏hepcidin在这一调控过程中可能起着重要作用.
