回交、DH和RIL偏分离群体遗传图谱的重新构建 被引量:3

摘要 分子标记偏离孟德尔分离比例(称偏分离)是一种普遍的生物学现象.虽然偏分离可能会影响标记间遗传距离的估计值和数量性状基因座(quantitative traitlocus,QTL)定位结果,但在遗传连锁图构建和QTL定位中偏分离的影响常常被忽视.在分子标记存在偏分离、显性和缺失情况下,根据隐马尔可夫模型,我们已发展了一种新的多点方法来重新构建F2群体分子标记连锁图,以解决偏分离对标记连锁图构建的影响.本文简化了上述方法以适用于回交、加倍单倍体和重组自交系群体.模拟研究表明:(ⅰ)两连锁偏分离基因座(segregation distortion locus,SDL)影响标记间遗传距离的程度随SDL遗传率和标记间遗传距离的增加而增加,但受样本容量的影响较小;(ⅱ)两连锁SDL一般会低估标记间遗传距离,但是,加性模型下两个SDL加性效应符号相反时会高估之,上位性模型下两SDL加性×加性效应为负时却不影响其估计;(ⅲ)用本文的方法均能矫正遗传距离估计值的偏差.将新发展的方法应用于已构建的一个存在严重偏分离的水稻(Oryza sativa L.)籼粳杂交组合IR28×大关稻的F7代重组自交系群体,重新构建了分子标记遗传图谱,并用Bootstrap法获得了遗传距离的95%置信区间.这些结果进一步印证了本文发展的新方法.这为数量性状和生活力性状的遗传分析提供了基础.为实际数据分析研制的DistortedMap计算机软件可供利用1).
