摘要 目的探讨缺氧诱导因子-1α(HIF-1α)和PTEN、P53在急性髓性白血病骨髓基质细胞中的表达及意义。方法分离培养28例急性髓性白血病患者和22例正常对照样本骨髓基质细胞,RT-PCR检测HIF-1αmRNA的表达,免疫组织化学技术检测基质细胞HIF-1α和PTEN、P53蛋白表达和分布。结果急性髓细胞白血病(AML)组HIF-1αmRNA和蛋白表达率显著高于对照组(P<0.05),AML组P53蛋白表达率显著高于对照组(P<0.05),而PTEN表达率显著低于对照组(P<0.01)。HIF-1α在骨髓基质细胞的阳性表达与P53的高表达和PTEN的低表达相关联(P<0.05)。结论HIF-1α可能是白血病异常骨髓造血微环境形成过程中的重要调控因子,HIF-1α的表达可能与PTEN和P53基因表达调控相关。 Objective To explore the expression and significance of hypoxic inducible factor-1α (HIF-1α ), PTEN, and P53 in stromal cells of acute myeloid leukemia (AML). Methods Stromal cells of 28 AML patients and 22 controls were isolated from bone marrow and cultured by adherent culture method. The expression of HIF-1α mRNA was detected by RT-PCR. The expression and distribution of HIF-1α, PTEN, and P53 proteins were detected by immunohistoehemical method.Results The expression level of HIF-1α mRNA and the positive rate of HIF-1α protein in AML group were higher than those of the control group ( P 〈 0.05). The positive rate of P53 protein was higher than that of the control group( P 〈 0.05), while the positive rate of PTFN protein was lower than that of the control. HIF-1α protein expression of stromal cells was associated with high P53 expression and low PTEN expression. Conclusion HIF-1α may be the important regulating factor of leukemic bone marrow And the expression of HIF-1α may be associated with the gene PTEN and P53 .
