摘要
目的:分离胃癌蛋白质组,找出胃癌组织的差异蛋白质点,为进一步鉴定胃癌的差异蛋白质打下基础。方法:利用二维聚丙烯酰胺凝胶电泳(two-dimensional polyacrylamide gel electrophoresis,2-DE)分离胃癌组和正常胃粘膜对照组总蛋白质,并通过优化的计算机图像分析软件分析电泳图谱。结果:在胃癌组织中点检测共有蛋白质点1 147个,正常胃粘膜组织中点检测共有蛋白质点1 079个,发现两者间共有164个差异蛋白质点,其中41个点仅在胃癌组织中表达,27个点仅在正常胃粘膜中表达,39个点在胃癌组织中高表达,57个点在胃癌组织中低表达。结论:通过优化的双向凝胶电泳和图像分析技术对胃癌蛋白质组进行研究,获得了胃癌蛋白质组与正常对照组的差异蛋白质点。为进一步利用质谱分析技术做差异蛋白质鉴定奠定了基础。
Objective:our purpose was to sieve the discrepancy protein from proteome and lay a foundation for verifying the particular protein sites in gastric carcinoma. Methods: 2-DE(two-dimensional polyacrylamide gel electrophoresis) was used to sever the general protein from tumor tissue and normal tissue, then analysed the results by computer-graph analysis software. Results: we verifed 1,147 protein spots in gastrica carcinoma tissue and 1,079 ones in normal specimens. Among them, there were 164 discrepancy protein spots. We found that 41 spots only emerged in gastric carcinoma and 27 ones in normal mucous. Meanwhile, 39 spots frenquently emerge and 57 ones were less expressed in tumor tissue. Conclusion: we got the discrepancy protein spots and laid hard basis for deeper research of discrepancy protein.
