三氯化铝改良牛心包瓣处理的研究

摘要 通过应用三氯化铝(AlCl_3)对戊二醛(Glutaraldehyde,GA)处理的牛心包瓣进行改良处理,探讨处理后牛心包力学特性及其在大鼠皮下钙化情况。方法:以AlCl_3对GA处理的牛心包瓣改良处理为实验组,以单纯GA处理(GA组)和未处理(PBS组)作对照。测定各组心包热收缩温度和各力学指标,并将各组心包片植入大鼠皮下分析钙化。结果:AlCl_3改良处理组热收缩温度高于GA处理组心包,但无显著性差异(P>0.05)。AlCl_3改良组各力学指标与GA组比较无显著差异(P>0.05),AlCl_3改良处理极显著抑制钙化发生(P<0.01)。结论:AlCl_3改良处理GA处理的牛心包瓣可提高热收缩温度,不影响其力学特性,且具良好的抗钙化性能。 This study was designed to assessed the ability of modified treatment with AlCl_3 (0.1mol/L) solution to inhibit the calcification of glutaraldehyde-fixed bovine pericardial valves. Bovine pericardial valves were treated in three methods. 1. pericardial valves were commercially treated with glutaraldehyde (GA) 2. valves were treated with GA and 0.1M AlCl_3 solution 3. fresh pericardial valves without any treatment. The mechanical properties of the bovine pericardial valves were tested. The calcium and aluminum content of the valves were determined in the 7, 21, 60, 90th days after the treated valves having been implanted subcutaneously in rats. Pathologic changes were examined by light and electron microscope. The mechanical Properties and shrinkage temperature of the valves treated with GA and AlCl_3 showed no significant difference from GA-fixed valves (P>0.05). Modified treatment with AlCl_3 could significantly inhibit the calcification of the bovine pericardial leaflets (P<0.01, compared with GA valves). The content of Al^(3+) did not loss from the valves treated with AlCl_3 solution. Modified treatment with 0.1M AlCl_3 solution can markedly inhibit the calcification and do not compromise the mechanical properties of the GA-fixed valves.
