骨形态发生蛋白在闭合性长骨骨折愈合中的作用 被引量:11

Immunohistochemical investigation of bone morphogenetic protein (BMP) in external callus of closed tibial fracture

摘要 为观察骨形态发生蛋白在闭合性长骨骨折愈合中的作用,以64只健康SD大鼠为动物,制备闭合性胫骨骨折动物模型,分别于12小时、1、3、5、7、9、14、28天取材,行恒冷切片,PAP染色,设立正常及PBS阴性替代对照。第3,5天纤维肉芽组织中的成纤维细胞样间充质细胞及第14天的新生骨小梁骨基质、成骨细胞、软骨细胞以及骨小梁间纤维组织内的成纤维细胞样间充质细胞为阳性反应,表明创伤使内源性BMP呈阶段性合成与释放,并诱导周围软组织以软骨内成骨方式向软骨及骨组织分化,形成外骨痂,参与骨折修复。 Sixty-four Sprague-Dawley rats were used. Closed fractures were produced manually in the middle of the shaft of tibia after intramedullary fixation. Eight animals each were killed at 12 hours, 1, 3, 5, 7, 9, 14, and 28 days after operation. The external calluses were dissected with the surrounding tissue. The specimens were cut with use of a cryostat. The slides were stained with peroxidaseanti-peroxidase (PAP) method. The control groups included normal muscle surrounding tibia and negative PBS-substitute. The immunohistochemical staining revealed that from day 3 to 5, the fibroblast-like mesenchymal cells in fibrous tissue positively reacted to bBMP-McAb. On day 14, when the cartilage was calcified into woven bone, the positive sig- hal were appeared in newly formed chondrocytes and fibroblast-like mesenchymal cells in fibrous tissue enclosed in newly formed trabecula. The control groups showed negative reaction. It revealed that trauma triggered off the synthesis and secretion of BMP, just before chondrogenesis,before and with osteogenesis.
