摘要
为探讨简便、实用、长期培养肝细胞的有效方法,以cytodex 3为材料,对Wistar大鼠肝细胞进行微载体粘附培养。结果发现:分离的肝细胞经限制贴壁、定期旋转后粘附于被覆胶原的微载体上呈浮游生长。存活并保持正常形态与白蛋白分泌功能平均达1月之久。因此,用微载体粘附方法可进行肝细胞的长时间、高活性及高密度培养。在肝细胞移植、生物人工肝等领域的研究中具有一定的实用价值。
Hepatocytes of Wistar rats were employed for attached culture by cytodex 3 microcarriers. After limiting attachment to dish and stirring at regular intervals, The cultural hepatocytcs were attached to collagencoated microcarriers survived and mainained the normal morphology and albumin secret function over one month. This method can be used as the long-time, highdensity and activity culture of hepatocytes and will suitable for the researches of hepatocyte transplantation and bioartificial liver.
