周期性机械拉伸对人肺上皮细胞增殖及粘弹性的影响

摘要 采用FX-4000基底加载系统对体外培养人肺上皮细胞A549施加周期性拉伸应变,通过流式细胞仪和微管吸吮技术,研究了不同波形的周期性机械拉伸对A549细胞的增殖及粘弹性的影响。结果表明:分别采用正弦波和方波,对A549细胞施加15%拉伸应变、频率30次/min周期性机械拉伸4 h,与对照组相比,正弦波拉伸对A549细胞增殖无显著影响,细胞的粘弹性参数k1和k2值有所减小,μ值无统计学差异。方波拉伸在显著抑制A549细胞增殖同时,细胞的粘弹性参数k1,k2和μ值均明显减小。从力学的角度证实了细胞骨架参与细胞生长的过程。 The proliferation and viscoelastic properties of human alveolar epithelium cells A549 were determined by using Flow Cytometer and micropipette aspiration technique respectively. In our studies, amplitudes of 15% strain were used. Stretch was applied in sinusoidal and square fashions at rate of 30 cycles/min for 4h. Our results showed that in sine wave groups, cyclic stretch had no effect on the proliferation of A549 cells. Furthermore, cellular elastic coefficient k1, k2 decreased slightly and viscous coefficient μ had no distinct difference compared with controls. However, viscoelastic parameters k1, k2 and μ decreased significantly in square wave groups compared with controls. Interestingly, the proliferation of cells A549 in square waveform was inhabited at the same time. It indicated that the cytoskeleton may participate in the process of cellular proliferation.
