摘要
目的:探讨蒙药肉蔻五味丸对心肌梗死后左室重构大鼠心肌c-Myc表达的影响。方法:SD大鼠96只,随机分为正常对照组、心肌梗死模型组、卡托普利治疗组、蒙药治疗组。其中蒙药治疗组又分低、中、高剂量组。模型组和各治疗组大鼠腹腔注射异丙基肾上腺素(ISO),卡托普利组和蒙药组分别给予卡托普利50mg/kg.d、低剂量肉蔻五味丸0.2g/kg.d、中剂量0.5g/kg.d、高剂量1g/kg.d灌胃,至各相应时段。称重并计算以重量指数,光镜下观察心肌病理改变,免疫组化法测定心肌原癌基因产物c-Myc的表达。结果:模型组大鼠左室重量指数比正常对照组升高,心肌c-Myc表达在各时段比对照组明显增加。而卡托普利组和蒙药组与模型组相比,左室重量指数、c-Myc表达不同程度降低。结论:蒙药肉蔻五味丸可降低心肌梗死后左室重构大鼠心肌c-Myc的表达,可能是其干预治疗心室重构的机理之一。
Objective:To study effects of Mongolian Drug RouKouWuWei Pill on expression of c- Myc in rats myocytes of ventricular remodeling following myocardial infarction. Methods: Ninety - six SD rats were randomly divided into normal control group,myocardial infarction group,Captopril group and Mongolian drug group, then the latter sepa- rated into three subgroups: low, mid and high dosage. The rats of myocardial infarction group and drug-treated groups were given isoproterenol(ISO), subsequently drug- treated groups given captopril 50mg/kg· d, low dosage Mongolian drug 0.2g/kg· d,mid dosage 0.5g/kg· d and high dosage lg/kg· d respectively until this experiment terminal. Left ventricular weight indexes were calculated and prom- oncogene translated prptein c - Myc was studied by immunoheistochemical staining. The pathologic change of myocardium was observed with microscope. Result: Left ventricular weight index and expression of c - Myc in the myocardial infarction group increased significantly compared with normal contml. The weight index and expression of c - Myc decreased in captopril group and Mongolian drug mid, high dosage group compared with myocardial infarction gruop. Conclusion:Mongolian Drug RouKouWuWei Pill can reduce the expression of c - Myc in rats myocytes of ventricular remodeling following myocardial infarction, which may be one of mechanisms on the ventricular remodeling following myocardial infarction.
