摘要
目的研究珍珠菜总黄酮苷(ZTF)的抗肿瘤作用及其机制。方法通过体内抑瘤试验,考察了ZTF对小鼠肝癌H22的影响;通过体外抑瘤试验,考察了ZTF对人白血病HL-60、K562细胞增殖的影响。并同时考察了ZTF对H22小鼠实体肿瘤组织PCNA表达的影响。结果ZTF对HL-60及K562细胞有明显的增殖抑制作用,ZTF400mg/kg对肝癌H22实体瘤有明显的抑制作用,抑瘤率在30%以上。ZTF可明显下调小鼠肝癌H22肿瘤组织中PCNA的阳性表达,其抑制强度亦呈剂量依赖性趋势。结论ZTF为珍珠菜的抗肿瘤作用有效部位,在体内外均具有一定的抗肿瘤活性,其机制可能是通过下调肿瘤细胞PCNA的表达,干扰肿瘤细胞DNA的合成。
Objective To study the anti-tumor effect and mechanism of total flavones (ZTF) exstracted from Lysimacbia cletbroides Duby. Methods Detect the effects of ZTF on transplant tumor models H22 in vitro, and its inhibitory effect on human leukemia cell HL-60 and K562 was observed. Then the infuence of ZTF on PCNA expression in transplant tumor H22 was studied. Results ZTF could significantly inhibit cell growth of HL-60 and K562; as to H22, the inhibition ratio was above 30% when treated with ZTF 400 mg/kg. ZTF could significantly reduce the expression of PCNA in the tumor of H22, and that was related with dosage. Conclusion ZTF is the effective extraction of Lysimachia clethroides Duby, which had anti-tumor effect in vivo and in vitro, and its mechanism is to interfere DNA synthesis in tumors by down-modulating the expression of PCNA.
