摘要
目的研究早期生长反应基因-1(Egr-1)在急性胰腺炎肝脏损伤机制中的作用。方法24只雄性Wistar大鼠随机均分为4组,分别胆管内注入生理盐水或不同浓度牛磺胆酸钠溶液,观察各组大鼠肝组织Egr-1免疫组化染色结果并进行胰腺病理评分,检测血清AST、LDH及TNF-α和IL-1β水平。从正常Wistar大鼠分离培养肝细胞,观察原代培养肝细胞在TNF-α刺激后Egr-1蛋白表达情况。结果肝组织Egr-1表达与急性胰腺炎肝损伤程度呈正相关,且原代培养肝细胞Egr-1蛋白表达水平与肝细胞损害程度呈正相关。结论Egr-1可能参与急性胰腺炎肝损伤过程,并且其作用机制部分依赖于细胞外信号调节激酶1/2(ERK1/2)。
Objective: To evaluate the role of early growth response gene- 1 (Egr- 1) in liver injury during rat tauroeholate - induced acute panereatitis model. Methods: Twenty - four male Wistar rats were divided into 4 groups. Normal saline was injeeted into eommon bile duet (CBD) of group A, and tauroeholate sodium solutions with different eoneentrations were injected into CBD of group B, C and D. Egr - 1 immunohistoehemistry staining and panereatie pathologie seoring were assessed, and the serum levels of AST, LDH, TNF-α and IL- 1β were measured.Egr- 1 protein expression by immunoblotting of primary hepatocyte culture stimulated with TNF-α and pretreated primary hepatocytes with PD98059 followed by TNF - α stimulation were also observed.Results: Egr- 1 expression of rat liver correlated with liver injury level during rat acute panereatitis, and high Egr- 1 protein expression of primary hepatocyte was observed when hepatocyte injury occurred. After pretreated with PD98059, TNF - α - stimulated hepatocytes showed a lower level of Egr- 1 protein expression compared with hepatocytes without pretreatment. Conelusions: Egr- 1 may play an important role in liver injury during acute panereatitis, and this effect is partly dependant upon extraeellular signal- regulated kinase 1/2 (ERK1/2) pathway.
