摘要
采用反相高效液相色谱(RP-HPLC)法,建立了大十桑椹的高效液相色谱指纹图谱。色谱条件为:Shimpack VP-ODS(250mm×4.6mm,5μm),流动相为0.05%磷酸-乙腈-四氢呋喃,梯度洗脱,流速为0.8mL/min,检测波长为313nm,柱温为30℃。结果检测出14个分离度良好的共有指纹峰,通过相关系数聚类分析及相似度计算,所有供试样品可归为一类,表明该方法可作为科学评价及有效控制桑椹及桑椹产品质量的可行的定性方法。
HPLC fingerprints and a sensitive and specific method for the quality control of the mulberry were studied. The gradient elution mode was applied in chromatographic separation. Shimpack VP-ODS (250mm× 4.6mm, 5μm) was used with the mobile phase: 0.05 %phosphoric acid water solution-acetonitriletetrahydrofuran, flow rate: 0.8 mL/min, detecting wavelength: 313 nm and the column temperature: 30℃. Ten samples collected from different areas were determined. The clustering analysis by DPS6.2 was used and the software of similarity analysis was applied for datum analysis. These 10 samples were classified as 1 cluster. The samples' quality was assessed by Similarity Evaluation System for Chromatographic Fingerprint of TCM 2004, and this method had a good repeatability and reproducibility. The method can be used to identify and evaluate the quality of Mulberry conveniently.
