转移性骨肿瘤中COX-2,VEGF及微血管密度的关系 被引量:4

Expressions of COX-2 and VEGF in metastatic bone tumor and their relationship with microvessel density

摘要 目的:探讨转移性骨肿瘤组织中环氧化酶-2蛋白(COX-2)、血管内皮生长因子(VEGF)的表达及微血管密度值(MVD)三者的相关性,以明确对转移性骨肿瘤实施临床干预的可能性.方法:采用免疫组化S-P法检测42例转移性骨肿瘤手术标本COX-2,VEGF蛋白及CD34的表达,根据CD34染色结果应用OLYMPUS-VANOX多功能显微镜,采用DMCⅡ型图像分析系统,测定计数MVD值,并同时取原发性恶性骨肿瘤、良性骨肿瘤标本作为对照,探讨COX-2和VEGF在转移性骨肿瘤的表达情况,以及转移性骨肿瘤组织中MVD值.结果:COX-2及VEGF在转移性骨肿瘤组的阳性表达率分别为62%和81%,MVD在转移性骨肿瘤组和原发性骨肿瘤组中的阳性表达率均高于良性骨肿瘤组,差异有统计学意义;转移性骨肿瘤组中表达率虽高于原发性骨肿瘤组,但差异没有统计学意义.提示MVD与骨肿瘤的发生与转移有密切的相关性.结论:COX-2,VEGF在骨肿瘤转移、发展过程中可能起重要作用;MVD可作为判断肿瘤恶性程度的客观指标. AIM: To study the expressions of cyclooxygenase-2 (COX-2) , vascular cell growth factor (VEGF) and CD34 and their correlation in metastatic bone tumor and to explore the feasibility of clinical intervention. METHODS: The expressions of COX-2, VEGF and CD34 were studied by immunohistochemical SP method in 42 specimens of metastatic bone tumor, 10 of benign bone tumor and 13 of osteosarcoma. The microvessel density (MVD) was assayed by counting CD34 positive blood vessels under microscope and with DMCII image analysis system. RESULTS: The positive rates of COX-2 and VEGF in metastatic bone tumor were 62% and 81% , respectively. The MVD values in metastatic group and osteosarcoma group were higher than that in benign bone tumor group ( P 〈 0.05 ), and metastatic group was higher than osteosarcoma group in MVD value, but there wasn't significant difference (P 〉 0. 05 ). CONCLUSION: COX-2 and VEGF might play important roles in the angiogenesis in human metastatic bone tumor, and MVD can serve as an objective index for judging the degree of rumor malignancy.
