摘要
目的探讨外源性碱性成纤维细胞生长因子(bFGF)对新生鼠缺氧缺血性脑损伤(HIBD)后巢蛋白(nestin)表达的影响。方法通过7日龄新生Wistar大鼠制作HIBD模型,设置对照组、治疗组,并设立假手术组。免疫组化方法观察巢蛋白表达的变化。结果各时点对照组巢蛋白阳性细胞数较假手术组增加,治疗组巢蛋白阳性细胞数较对照组增加明显,各时点三组比较差异有统计学意义(P<0.01)。结论新生鼠HIBD后脑缺血易损伤区巢蛋白表达增加,可能与脑损伤后神经再生有关;给予外源性bFGF可增强新生鼠脑缺氧缺血后中枢神经系统巢蛋白的表达,在神经细胞损伤的修复中发挥一定的保护作用。
Objectives To investigate the effect of exogenous basic fibroblast growth factor (bFGF) on brain nestin expression after hypoxic-ischemic brain damage (HIBD)in neonatal rat. Methods Eighty HIBD models of neonatal Wistar rats were made by shearing right arteria carotis eommunis and then breathing 8%O2+92%N2 for two hours. The models were divided into two groups randomly: the trial group of bFGF and the control group. Other forty rats were taken into the sham operation group. Expressions of nestin were examined with immunohistochemical staining at the 4th, 7th, 10th, 17th and 24th days after the operation. Results The number of nestin-positive cells in hippocampal CA1 region was the highest in the trial group, higher than in the control group, in comparison with the sham operation group, there was a significantly difference in three groups at all points (P 〈 0.01). Conclusions 1.The expression of nestin increased in the brain of neonatal rats with HIBD, it might indicate the regeneration of neuroeytes after HIBD. 2.Exogenous bFGF could enhance the expression of nestin, which may play an important role in restoration of the neurons damage after hypoxia-ischemia.
