摘要
目的探讨细胞间黏附分子(ICAM)-1和淋巴细胞功能相关抗原(LFA)-1在干燥综合征(SS)的发病机制中的作用。方法用免疫组织化学SABC法,3,3-二氨基联苯胺(DAB)显色,对32例不同程度干燥综合征患者唇腺组织和15名正常对照组进行了ICAM-1和LFA-1的检测。结果①ICAM-1和LFA-1在SS组的表达明显高于对照组(P<0.01)。ICAM-1分布广泛,以导管上皮表达最强,其他腺上皮、血管内皮及淋巴细胞浸润区均有表达:LFA-1则主要表达于淋巴细胞浸润区。②ICAM-1及LFA-1在SS的表达随着淋巴细胞浸润程度的加重呈递增趋势。③ICAM-1和LFA-1呈直线相关(r=0.989,P<0.01)。结论上皮细胞在SS中起非专职抗原提呈细胞作用,积极参与了诱导和维持淋区细胞的浸润。
Objective To explore the role of intercellular adhesion molecule-1 (1CAM-1) and lymphocyte function-associated antigen-1(LFA-1 ) in the pathogenesis of Sjoigren's syndrome (SS) and provide a theoretical basis for clinical therapy. Methods hnmunohistochemieal method was used to detect these two cellular adhesion molecules in labial salivary glands of primary Sjogren's syndrome patients and 15 healthy controls. Semiquantitative analysis was performed by image analysis software. Results ①In salivary gland samples, the expression of both 1CAM-1 and LFA-1 was significantly higher compared to that of controls (P〈 0.01 ), ICAM-1 expression was detected mainly in ductal epithelial cells and glandular epithelial, myoepithelial, endothelial of SS patients, while LFA-1 expression was expressed in lymphocytes. ②There was a positive correlation hetween lymphocyte focus score and ICAM-1 expression (P〈0.01) as well as LFA-1.③ Linear correlation between expression of ICAM-1 and LFA-1 was detected. The correlation coefficient was r=0.989 (P〈0.01). Linear regression equation was Y=1.037X+4.612×10^-4. Conclusion The high expression of 1-CAM-1 in the salivary gland epithelial cells indicats that they act as nonprofessional APCs, these cells actively paticipate in the induction and maintenance of lymphocyte infiltration. The ICAM-1/LFA-1 pathway may have an important role in the pathogenesis of SS.
