CpG ODN107增强人脑胶质瘤细胞放射敏感性的研究 被引量:7

摘要 目的探讨CpG ODN107增加人脑胶质瘤细胞(CHG-5)放射敏感性的作用及可能机制。方法用不同浓度CpG ODN107处理CHG-5细胞,然后经放射线照射,MTT法检测细胞的增殖情况,体外培养集落形成率与细胞划痕法观察细胞的集落形成和迁移能力,ELISA法检测细胞TNF-α的分泌水平。结果CpG ODN107抑制CHG-5细胞的增殖,并呈量效关系。CpG ODN107(10μg/mL)联合放射线照射,抑制CHG-5细胞的增殖作用最强(0.52±0.76)。CpG ODN107(10μg/mL)同样能抑制CHG-5细胞增殖和迁移,并且增加了TNF-α的分泌,在联合照射后作用更加显著(193±0.58)pg/mL。结论CpG ODN107对CHG-5有明显的放射增敏作用,其机制可能与CpG ODN107增强肿瘤细胞TNF-α的分泌有关。 OBJECTIVE: To investigate the influence of CpG ODN107 on the radiosensitivity of human glioma cell line (CHG-5) and its mechanism. METHODS: CHG-5 was exposed to CpG ODN107 at different concentrations or/and irradiation, the abilities of proliferation were measured by MTT test, the abilities of colony formation and migration were observed by the colony formation test and scratch method, the TNF-α secretion by CHG-5 was detected by ELISA. RESULTS: CpG ODN107 dose-dependently inhibited the proliferation of CHG-5.and the inhibitory action was enhanced(0.52±0.76)when CpG ODN107(10μg/mL)was combined with β-ray irradiation.The colony formation and migration of CHG-5 were also inhibited by CpG ODN107(10μg/mL)or combined with irradiation ,with the increase of TNF-α seceretion by cells (193±0.58)pg/mL.CONCLUSION:Cpg ODN107 can increase the radiosensitivity of human glioma cell line (CHG-5),which may be associated with enhancing TNF-α secretion by cells.
