摘要
目的探讨不同分化程度肝癌组织中β-G mRNA的表达及其意义。方法采用逆转录-聚合酶链反应(RT-PCR)方法,对10例正常肝脏及38例不同分化程度肝癌组织中β-G mRNA的表达进行对比研究。结果β-G mRNA在正常肝和肝癌组织中均有表达,且表达产物大小一致,为422bp,半定量相对表达含量为正常肝组织(1.71±0.32)与高分化肝癌(2.17±1.07)比较,差异无统计学意义,t=2.513,P=0.264;而与中分化肝癌(3.67±1.27)和低分化肝癌(5.63±1.62)比较,差异有统计学意义,t=6.721,P=0.036和t=11.472,P=0.0067。结论β-G mRNA在不同分化程度肝癌组织中的表达差异有统计学意义,随着恶性程度的增加而表达量呈递增趋势,提示β-G mRNA可能参与了肝细胞的癌变过程,对β-G mRNA的进一步探讨,可能对肝癌的分子机制研究有重要意义。
OBJECTIVE:To explore the expression and significance of -glucuronidase (β-G) mRNA in different differentiation tissues of hepatocellular carcinoma. METHODS: Reverse transcription polymerase chain reaction (RT-PCR) technique was used to detect the expression of β-G mRNA in 10 cases of normal liver tissue and 38 cases of different differentiation tissues of hepatocellular carcinoma. RESULTS: β-G mRNA was expressed in the normal liver tissue and in hepatocellular carcinoma, the size of their products were the same, 422 bp, and the opposite semiquantitative expressive contents were (1. 71 ±0. 32) in normal liver tissue and (2. 17±1.07) in hight differentiation hepatoeellular carcinoma, respectively, without statistical significant difference, t = 2.513, P =0.264. However, there were signifiant differences between the normal liver tissue and middle differentiation hepatocellular carcinoma (3.67 ± 1.27) or low differentiation hepatocellular carcinoma (5.63±1.62), t=6.721, P=0.036 or t=11.472. P=0.006 7. CONCLUSIONS: Expression of β-G mRNA is remarkable different among different differentiation hepatocellular carcinomal tissues, and and with malignant degree increases, the quantity of the expression increases progressively. It points out that β-G mRNA may participate in the course of carcinomal changes of liver tells, and it is important to explore further β-G mRNA for molecular mechanism of hepatocellular carcino ma.
