尿液双向电泳蛋白质样品的制备

摘要 尿液的获取简便易行、没有侵入性,尿液中的蛋白质能反映机体,特别是肾脏的功能,使尿蛋白质组学研究成为大家关注的一个焦点。双向电泳技术具有高通量、半定量以及能同时给出蛋白质分子量、等电点及修饰等信息的优点,吸引着人们利用该技术做了大量尿蛋白质组学研究工作。然而尿液中蛋白质成分较复杂且含量低、并受到众多不确定的修饰和降解等因素的影响使尿蛋白质样品的制备比较困难。本文综述了尿中蛋白成分和目前用于双向电泳的尿蛋白提取方法以及为了获得高分辨率双向电泳图谱而对蛋白样品进一步的处理方法,希望能给读者以启示。
