海洛因依赖大鼠部分脑区超微结构变化研究 被引量:5

摘要 目的探讨海洛因依赖大鼠中枢神经系统毒理病理损伤机制。方法海洛因依赖组(实验组)按逐日递增原则皮下注射海洛因建立海洛因依赖模型,对照组注射等量生理盐水。透射电镜观察海洛因依赖组及对照组部分脑区:兰斑(LC)、中脑导水管周围灰质(PAG)、杏仁核、黑质和海马区超微结构变化。结果海洛因依赖大鼠部分脑区神经细胞胞体固缩,神经纤维水肿,有髓神经纤维髓鞘松解,胞内线粒体肿胀、嵴断裂消失,线粒体畸形,内质网扩张、脱颗粒,多聚核糖体解聚,轴突内可见微囊形成,部分轴突内细胞器稀疏。结论海洛因依赖对中枢神经系统具有损害,相关脑区神经细胞和神经纤维超微结构皆发生不同程度改变。 Objective To discuss the central nervous system toxicologypathology damage mechanism in the heroindependent rats. Methods Heroin was injected according to the princicle of daily increasing dose to establish heroindependent models of rats. Saline of equal dose was administrated in control group.To observe the ultrastructure changes of locus coeruleus, periaqueductal gray, substantia nigra, corpus amygdaloideun and hippccampus in the heroin dependence group and the control group rats by transmission electron microscope. Resulls In the heroin-dependent rats, the nerve cells were dropsied or pyknosised, neuropil was partly edematous, myelin of myelinated nerve fibers was of brisement, mitcchondria were swelled and deformed,endoplasmic reticulums were dilated,polyribocomes were disaggregated, axons and dentrites were partly dropsied, organelles were rarefied, synaptic vesicles were accumulated in some synaptic terminals. Five cerebral regions were normal in the rats of the control group. Conclusions The central nervous system was damages in the heroin-dependent rats , in partial cerebral regions,the nerve cell and the nerve fiber uhrastructures had the varying degree changes, heroin may result ischemic-anoxic and degradation changes in nerve cells.
