大黄对多发性创伤兔血中细胞因子变化的影响 被引量:2

摘要 目的观察多发性创伤兔血中肿瘤坏死因子α(TNF-α)、白细胞介素-6、8(IL-6、8)变化及探讨大黄的治疗和保护作用。方法清洁级大白兔18只,复制多发性创伤兔的模型,随机分为三组:治疗组(常规治疗+大黄治疗6只),对照组(常规治疗+安慰剂治疗6只),正常组(6只)。大黄治疗组和对照组分别在创伤前0min及创伤后30min、60min、120min、240min、480min时间点采血测定血浆TNF-α、IL-6、8的水平,正常组记录相应时间点测TNF-α、IL-6、8的水平。结果治疗组和对照组多发性创伤后TNF-α、IL-6、IL-8的水平逐渐升高比正常组的相同时间点有差异(P<0.05),治疗组与对照组创伤后4h水平有差异(P<0.05),创伤后8h有显著差异(P<0.01);而治疗组其升高程度显著低于对照组(P<0.01)。结论THF-α、IL-6及IL-8参与了多发性创伤后的病理生理过程,应用大黄可降低血浆中TNF-α、IL-6及IL-8的水平,明显减轻机体的全身炎症反应程度。 Objective To observe the level changes of tumor necrosis factor alpha(TNF-α),intedeukin-6(IL-6),interleukin-8 (IL-8) in plasma of the rabbits of seriously multiple traumatic and determine the effect of Rhubarb on these indexes. Methods Eighteen rabbits were randomly divided into 3 groups, namely treatment with Rhubarb group (treatment group,n= 6),treatment with normal saline group (control group, n=6),normal group (n=6).In treatment group and control group to measure TNF-α,IL-6 and IL-8 of plasma at pre-seriously multiple trauma,0.5 hour,1 hour,2 hours,4 hours and 8 hours after seriously multiple trauma. In normal group TNF-α,IL-6 and IL-8 of plasma were also measured in corresponding to courses of seriously multiple trauma. Results There were significant difference about the levels of plasma TNF-α,IL-6 and IL-8 of treatment group and control group in rabbits each time point after seriously multiple trauma vs nomlal group all increased (P〈0.05). there were significant difference about the levels of plasma TNF-α,IL-6 and IL-8 of treatment group and control group of 4 and 8 hours after seriously multiple trauma (P〈0.05),but levels of treatment group vs control group very lower (P〈0.01).Condusions It is suggested that TNF-α,IL-6 and IL-8 may play a role in pathophysiology after seriously multiple trauma. The application of Rhubarb can decrease the levels of plasma TNF-α,IL-6 and IL-8 of after seriously multiple trauma. Its pharmaceutical effects may function through gastrointestinal mechanisms.
