宽心合剂对大鼠急性心肌梗死后心室重构作用影响的研究 被引量:2

摘要 目的探讨宽心合剂对大鼠急性心肌梗死(AMI)后心室重构作用的影响。方法取健康雄性SD大鼠24只,随机分为假手术组(C组)、AMI模型组(M组)和宽心合剂治疗组(K组),每组8只。除C组只穿线不结扎外,其余2组采用开胸结扎左冠脉前降支的方法制作大鼠AMI模型。术后12 h开始,K组给予宽心合剂14 mL/(kg.d)灌胃,其余2组在相应时间点给予清水10 mL/(kg.d)灌胃。术后4周,断头处死各组大鼠,测定大鼠体质量(mB)、心脏质量(mH)、左室质量(mLV)、心脏质量/体质量(mH/mB)、左室质量/体质量(mLV/mB)及心肌胶原含量,并分别采用MASSON染色光镜下观察心肌胶原增生情况。结果术后4周M组大鼠心肌肥厚指数和心肌胶原含量较假手术组显著升高(P均<0.01),K组大鼠上述指标显著低于AMI模型组。MASSON染色结果:M组胶原纤维明显增生,经宽心合剂治疗后心肌胶原纤维堆积均较M组明显减轻。结论宽心合剂有明显抑制大鼠AMI后心肌纤维化作用,但不能完全阻止其发生。 Objective To investigate the effect of herbal compound Kuanxin on ventricle remodeling in rats after acute myocardial infarction. Methods Twenty - four male healthful SD rats were divided into three groups randomly. Each group included eight rats, namely: sham operation control group (group C), AMI model group (group M), and herbal compound kuanxin group (group K). Models of ligation of the anterior descending branch of left coronary artery were established. In the group C, the heart was exposed after the chest had been opened, but not ligated. All rats were keep on fed for four weeks after the operation. Each rat in group K was fed with herbal compound Kuanxin 14 mL/(kg·d) in addition. At the end of the experiment, all rats were killed. Heart tissue were collected for being measured body weight ( m B ), heart weight ( mH ), left ventricle weight ( m LV), mH / m B , m LV/ m B and collagen contents of myocardial. Heart tissue were collected for being observed by MASSON dyeing under light microscope in each group. Results Four weeks after operation, the content of myocardial collagen and myocardial hypertrophy indexes in all model established groups were significantly increased than those in group C (all P 〈 0.01 ). The two indexes in group K were significantly decreased compared with group M (all P 〈 0.01). The results observed under light microscope by MASSON dyeing showed that, there were less myocardial collagen fibers in drug intervene groups than those in group M. Conclusion Herbal compound Kuanxin has obvious effect in suppressing restrain ventricle remodeling after AMI in rats, but can not stop it.
