淫羊藿、秦皮醇提取物体外抗乳腺癌细胞增殖的研究

Antiproliferative Activities of Alcohol Extracts of Herba Epimedii and Cortex Fraxini on Breast Cancer Cell Proliferation: in Vitro Study

目的：考察淫羊藿和秦皮的不同浓度乙醇提取物体外抗乳腺癌细胞增殖的作用.方法：将淫羊藿和秦皮20%、40%、70%和95%乙醇提取物作用于人乳腺癌MCF 7（ER^＋）和MDA-MB-231（ER^-）细胞,用MTT法检测其抗增殖活性.结果：淫羊藿95%的乙醇提取物在100～800 μg·mL^-1浓度范围内均能显著抑制人乳腺癌MCF-7细胞的增殖,70%的乙醇提取物也显示了一定的抗增殖活性,而20%和40%的乙醇提取物未见明显的抗增殖作用;但对人乳腺癌MDA-MB 231细胞,淫羊藿不同浓度的乙醇提取物均无明显的抗增殖作用.秦皮95%和70%的乙醇提取物在50～400 μg·mL^-1浓度范围内均能显著抑制人乳腺癌细胞的增殖,而40%和20%的乙醇提取物未见明显的抗人乳腺癌细胞增殖的作用.结论：淫羊藿和秦皮的乙醇提取物有一定的抑制乳腺癌细胞增殖的作用.

OBJECTIVE： To conduct an in vitro study on the antiproliferative activities of different concentrations of alcohol extracts of Herba Epimedii and Cortex Fraxini on human breast cancer cell proliferation. METHODS： The human breast cancer MCF-7（ER^＋） cells and MDA-MB 231（ER^-） cells were incubated with 20%, 40%, 70% and 95% alcohol extracts of Herba Epimedii and Cortex Fraxini, with the antiproliferative activities of the alcohol extracts determined by MTT assay. RESULTS： The 95% alcohol extract of Herba Epimedii could significantly inhibit the proliferation of human breast cancer MCF-7 cell lines at the doses of 100-800 μg·mL^-1; the 70% alcohol extract also showed a certain antiproliferative activity, whereas the 20 % and 40% alcohol extracts showed no significant antiproliferative activity. Four concentrations of alcohol extracts of Herba Epimedii showed no significant antiproliferative activity on human breast cancer MDA-MB-231 cells. The 95% and 70% alcohol extracts of Cortex Fraxini could significantly inhibit the proliferation of human breast cancer cell lines at the doses of 50-400μg·mL^-1 but no significant antiproliferative activities of the 40% and 20% alcohol extracts on human breast cancer cells were noted. CONCLUSION： The alcohol extracts of Herba Epimedii and Cortex Fraxini have a certain inhibition activity on the growth of the breast cancer cells in vitro.
