摘要
目的探索海藻糖(trehalose)作为封闭液用于酶联免疫吸附检测(enzyme linked immunosorbent assay,ELISA)实验系统的可行性。方法在不同的抗原抗体反应体系采用不同浓度的海藻糖作为封闭液,对直接、间接及双抗体夹心ELISA敏感性与检测本底的影响。结果在一定的封闭时间里,不同浓度的海藻糖封闭液均可以在不影响ELISA反应体系敏感性的前提下增强对于非特异性吸附的抑制,封闭效果明显优于或等同于其他传统封闭液以及二糖类封闭液;结论海藻糖能够作为封闭液用于ELISA,并有望替代部分ELISA体系中的传统封闭液。
Objective To estimate the possibility of applying trehalose as blocker in enzyme linked immunosoibent assay (ELISA) .Methods Trehalose blocking reagent of different concemtration and blocking time in the direct-, indlrect-and sandwieh- ELISA system were tested to observe the influence on sensitivity and non - specific absorption comparing with some conventional blocking reagents. Results Trehalose blocking buffers of different concemtration are all able to decrease non-specific absorption while without affecting the sensitivity of the ELISA systerm during 2 hours, which are much better than or as well as some other blocking buffers other disaccharides blocking reagents. Conclusion Trehalose blocking reagent can be used in ELISA, which will be expected to replace some conventional blocking solutions.
