摘要 目的探讨不同浓度全反式维甲酸(ATRA)对食管癌细胞系EC109的生长抑制作用及其机理。方法不同浓度ATRA作用于细胞EC109,采用MTT法检测细胞生长;通过流式细胞仪检测和DNA梯状电泳证实凋亡存在;用RT-PCR方法检测凋亡诱导过程中p53、bcl-2和bax基因表达变化。结果ATRA呈剂量及时间依赖性抑制细胞生长;琼脂糖电泳呈现凋亡DNA梯状条带;流式细胞仪分析存在明显的凋亡峰;ATRA处理组bax和p53基因表达上调,但bcl-2基因表达下调。结论ATRA具有抑制食管癌细胞株EC109生长作用,其机理可能是诱导细胞凋亡,其分子机制可能与bcl-2/bax和p53的表达相关。 Objective To study the effects of a wide range of concentration of all-trans retinoic acid (ATRA) on esophageal cancer cell EC109. Methods The cell growth activity was detected by MTT assay after treatment with different concentration; the apoptosis was detected by flow cytometry assay and DNA Gel electrophoresis; the expressions of p53 and bcl-2/bax mRNA were assessed by RT-PCR. Results The cell growth was depressed by ATRA depending on concentration and duration obviously; the presented apoptosis features were observed; DNA gel electrophoresis showed DNA ladder; flow cytometry analysis showed an obvious apoptosis peak; the expressions of bax and p53 were up-regulated while the expression of bcl-2 was down-regulated in the treated group. Conclusions ATRA inhibits the growth of esophageal cancer cell EC109 obviously through inducing cell apoptosis, its molecular mechanism may be related with the expressions of bcl-2/bax and p53.
