摘要
选择60只断奶湖羊母羔羊,分为6组,1-5组为免疫组,分别用pES/2SS+pE-CpG、pES/2SS+细菌DNA、pES/2SS-GMCSF、pES/2SS脂质体和pES/2SS免疫,第6组为对照组。结果显示,首次免疫4周和加强免疫2周后,细菌DNA佐剂组的抗生长抑素抗体水平显著高于pES/2SS单独免疫组(P〈0.05),其抗体阳性率也最高,达50%。比较全期14周免疫羊的体质量增加情况,最优为pE-CpG佐剂组,其次为细菌DNA佐剂组,分别比对照组高33.0%和31.6%(P〈0.01),且显著高于pES/2SS单独免疫组(P〈0.05)。加强免疫2周后,免疫组GH和IGF-Ⅰ水平显著高于对照组(P〈0.05),抗体阳性羊的GH和IGF-Ⅰ水平极显著高于抗体阴性羊(P〈0.01)。结果表明:几种佐剂配合pES/2SS免疫,产生了免疫佐剂效应,能增强生长抑素DNA疫苗的免疫效果。
Sixty female Hu lambs were divided into six groups to immunized with different adjuvant such as the pE-CpG plasm plasmid, E. coil DH5α, the crude liposome were combined with pES/2SS, pES/2SS-GMCSF, pES/2SS and the control group. The results showed that the level of antibody of immunized group was remarkably increased compared with the control ( P〈0.05 ). The antibody against SS in the E. coil DH5α group was remarkably higher than that in the pES/2SS alone at week 4 after primary immunization and week 2 after booster. The sheep immunized with the E. coil DH5α group got the highest rate of positive antibody (50%). During the whole experimental period (14 weeks) , body weight gains in the pE-CpG and the E. coil DHSαgroups were 33.0% and 31.6%, respectively, which were higher than that of the control (P 〈0.01 ) and pES/2SS alone (P 〈 0.05). Two weeks after the booster, the level of serum GH and IGF - Ⅰ in the immunized group were remarkably higher than those in the control (P 〈0.05), and those with positive antibody higher than negative antibody ( P〈0.01 ). These results suggest that different adjuvant are combined with pES/2SS, can produce immunity adjuvant affect, have an enhancement effect on the SS DNA vaccine.
