摘要 目的建立谷氨酸损伤模型并探讨重组人酸性成纤维细胞生长因子(rhaFGF)对谷氨酸损伤神经细胞的干预作用。方法利用流式细胞仪观察rhaFGF对正常培养的神经细胞细胞周期的影响,建立细胞损伤模型,将实验分空白组、模型组和rhaFGF保护组,通过形态观察以及MTT法检测各实验组的细胞活力。结果rhaFGF能促进DNA的合成,谷氨酸对细胞损伤程度随浓度的增大而增大,rhaFGF能够拮抗谷氨酸的神经毒。结论rhaFGF能促进神经细胞生长同时拮抗谷氨酸兴奋性毒性而发挥神经保护作用。 Objective To study the protective effects of rhaFGF on cultured neuron SH-SY5Y iniured by glutamate. Methods The neuron SH-SY5Y damaged model in vitro was established with the treatment of ecltatory amino acid; cell cycle was analyzed with flowcytometer; morphological change was observed with microscope. Cell survival status was measured with MTT method. Results rhaFGF promoted the DNA synthesis significantly; cellular damage increased with the treatment of increasing gluamate dosage, rhaFGF improved the survival rate of SH-SY5Y and inhibited the cell damage induced by excitatory amino acid. Conclusion rhaFGF could improve the survival rate of neurons.
