CFDA SE标记观察人成骨细胞与骨支架材料附着情况的实验研究

摘要 目的通过荧光活性染料CFDA SE标记观察人成骨细胞与nano-hydroxyapatite/collagen/Poly L-lactic acid(nHAC/PLA)骨支架材料黏附生长情况。方法用CFDASE标记人成骨细胞,接种于nHAC/PLA骨支架材料上,测定细胞的黏附率。于接种后第1、3、7天检测荧光光度,并用激光共聚焦显微镜观察细胞在支架材料上的生长状况。结果CFDASE标记的人成骨细胞的黏附率为(97.99±1.43)%,未标记的人成骨细胞黏附率为(98.57±1.17)%,两者无统计学差异(P>0.05);随时间增长,nHAC/PLA骨支架材料内的细胞数量增多,沿孔壁生长良好,但总荧光光度值未发生明显变化。结论荧光活性染料CFDASE标记不影响人成骨细胞在nHAC/PLA骨支架材料上的黏附,可作为观察人成骨细胞在nHAC/PLA骨支架材料生长情况的实验手段。 Objective To study the adhesion of CFDA SE labeled human osteoblast to nano-hydroxyapatite/collagen/Poly(L-lactic acid (nHAC/PLA) scaffold material. Methods Human osteoblasts were labeled with CFDA SE and then seeded on nHAC/PLA scaffold material.The adhesion efficiency was calculated after 24hrs' culture, with the labeled human osteoblast as a control .The cell-nHAC/PLA composite was observed under confocal laser scanning microscope on the 1 st, 3rd, and 7th days after seeding. Results The adhesion efficiencies of CFDA SE labeled and unlabeled human osteoblasts were (97.99 ±1.43)% and (98.57 ±1.17)%, respectively, and there was no significant difference between the test group and control group( P 〉 0.05) .The cells grew well along with the hole wall, but there was no significant difference on total fluorescence intensity after 7 days' culture. Conclusion Application of vital fluorescence dye CFDA SE to human osteoblasts shows no effect on the adhesive ability of MG-63 cells, and CFDA SE label can be used as a good method to observe the adhesion of human osteoblasts on nHAC/PLA scaffold material.
