摘要
目的研究外源性牛血浆纤维黏连蛋白(fibronectin,FN)对体外培养的大鼠成骨细胞凋亡的影响。方法采用体外培养技术原代培养大鼠成骨细胞,Western blotting法检测FN作用后成骨细胞凋亡相关基因Bcl-2及Bax的蛋白表达变化。结果体外培养的成骨细胞呈梭形、三角形或多角形,多角形细胞有多个矢状突起,核卵圆形常居一侧,Ⅰ型胶原免疫组化染色呈阳性。FN作用后细胞凋亡相关基因Bcl-2的蛋白表达增强;Bax基因的蛋白表达减弱。结论一定浓度的牛血浆FN能够抑制成骨细胞凋亡,其机制之一是增强大鼠成骨细胞胞浆内Bcl-2基因所表达的蛋白,同时减弱Bax基因的蛋白表达。
Objective To investigate the effects of bovine plasma fibronectin on the apoptosis of rat osteoblasts in vitro. Methods Cells were cultured. Western blotting was used to determine the effects of various concentrations of bovine plasma fibronectin on the expression of Bcl-2 and Bax protein of osteoblasts in vitro.Results Fibronectin can increase the protein expression of Bcl-2 and decrease the protein expression of Bax. Conclusions Certain concentrations of bovine plasma fibronectin may suppress apoptosis of rat osteoblasts in vitro, which attributes to the increase of the protein expression of Bcl-2 and decrease the protein expression of Bax.
