摘要
目的:阐明大青叶的中枢解热机制,为其临床应用提供实验依据和理论指导。方法:实验用新西兰兔复制白介素-1β(interleukin-1β,IL-1β)发热模型,在观察大青叶注射液(Daqingyeinjection,DQYI)解热作用的基础上,应用原位杂交技术检测静脉注射DQYI对IL-1β作用下新西兰兔视前区-下丘脑前部(preopticanteriorhypothalamus,POAH)组织中前列腺素E3受体(E-typeprostaglandinreceptor,EP3)mRNA表达的影响。结果:(1)DQYI静脉注射后对正常新西兰兔结肠温度没有明显影响,与生理盐水组相比,最大体温上升高度(△T)及1h体温反应指数(TRI1)两组间无统计学意义(P>0.05);IL-1β组在静脉注射IL-1β后,结肠温度逐渐升高,△T及TRI1与生理盐水组相比,也有统计学意义(P<0.01);而DQYI+IL-1β组结肠温度上升峰值明显低于IL-1β组,两组△T及TRI1比较,有显著性差异(P<0.05)。(2)新西兰兔下丘脑POAHEP3mRNA在生理状态下仅有少量或没有表达;IL-1β诱导发热后,EP3mRNA表达明显增强;而DQYI能减少IL-1β作用下EP3mRNA的表达(P<0.05)。结论:DQYI对IL-1β诱导的新西兰兔发热有解热作用;且其解热作用机制可能与其抑制下丘脑EP3mRNA的表达有关。
AIM: To study the antipyretic effect and mechanism of Daqingye injection ( DQYI ). METHODS: The IL-1β-induced febrile New Zealand rabbits were chosen as experimental models and the antipyretic effect of DQYI was observed. The expression of EP3 mRNA was investigated by using in situ hybridization (ISH) in POAH. RESULTS: First, the colonic temperature went up gradually after intravenous(i, v) IL-1β. The peak value of temperature(△T) and thermal response index (TRI1) in IL-1β-treated group were higher than those in control group ( P 〈 0.01 ). The AT and TRI, of in DQYI and IL-1β- treated group were lower than those in IL-1β-treated group ( P〈0. 05). The temperature of DQYI-treated group showed no distinguished difference compared with that in control group ( P 〉 0.05). Second, the expression of EP3 mRNA in the POAH of IL-1β-treated group increased markedly compared with that in control group ( P 〈 0.01 ). The expression of EP3 mRNA treated by IL-1β + DQYI in the POAH decreased strikingly compared with that in IL-1β group ( P 〈 0.05). CONCLUSION: DQYI has distinct antipyretic effect on IL-1β-induced fever. The mechanism might be the inhibition of EP3 mRNA expression in POAH from rabbits.
